CJC-1295 without DAC, also called modified GRF (1-29), is an analog of the first 29 amino acids of growth hormone-releasing hormone (GHRH), with four substitutions that increase its resistance to enzymatic degradation.

It acts on the GHRH receptor in the anterior pituitary, stimulating the release of growth hormone. Because it preserves the physiological release mechanism, it maintains the pulsatile pattern and the negative feedback by somatostatin, unlike direct administration of exogenous growth hormone.
The substitutions relative to native GHRH protect the enzymatic cleavage sites, especially against DPP-4, which prolongs its functional presence without extending it into the multi-day range that characterizes the DAC version.

GHRH and its analogs have an established pharmacological basis, and at least one analog of this family —tesamorelin— is approved as a medicine for a specific indication. CJC-1295 itself is not approved in any country.
In the United States it was placed in Category 2 of the 503A list in September 2023 and removed from that category in September 2024, after its nomination was withdrawn. Leaving Category 2 because a nomination was withdrawn is not a favorable evaluation: it is the interruption of the evaluation procedure.
