What is GHK-Cu?

GHK-Cu is a tripeptide with the sequence glycyl-L-histidyl-L-lysine complexed with divalent copper. It is one of the best-characterized peptides in skin biology, and one of the few in the catalog with an established cosmetic use and decades of commercial formulation behind it.

It was identified in human plasma by Loren Pickart in the 1970s, in the context of research on age-associated plasma factors. Its plasma concentration declines markedly with age, an observation that sparked much of the later interest in the compound.

In terms of chemical identity, it is classified as Copper tripeptide, its CAS number is 89030-95-5, its molecular weight is 403.94 g/mol. These identifiers are what allow the material received to be checked against the batch Certificate of Analysis.

Mechanism studied in preclinical research

Copper is not an inert companion: the GHK tripeptide has a high affinity for divalent copper and the complex acts as a physiological carrier of the ion. Much of the described activity depends on that delivery of copper to copper-dependent enzymes, including lysyl oxidase, which is involved in the cross-linking of collagen and elastin.

In dermal models, modulation of the expression of type I collagen, elastin, glycosaminoglycans and matrix metalloproteinases has been described, that is, of both the synthesis and the controlled degradation of the extracellular matrix. That dual effect is what is described as remodeling, and it distinguishes it from a simple synthesis stimulus.

What does the evidence on GHK-Cu show, and where does it end?

Within this catalog, GHK-Cu is one of the compounds with the most established topical dermatological evidence, with literature on wound healing and skin quality parameters. It is also the only case with a split regulatory status: in the FDA update of September 29, 2023 it was placed in Category 1 with limitations for topical use, while its injectable routes were flagged in Category 2.

That route-based distinction is exactly the nuance worth keeping in mind. Topical evidence does not automatically transfer to the injectable route, which is the one the FDA flagged and for which the data are much more limited.

The material in this catalog is a research reagent, not a finished cosmetic or a medicine. A commercial cosmetic with GHK-Cu is a formulated product, with validated stability, preservation and concentration, which is something else.

Handling, reconstitution and storage

The vial is delivered lyophilized, that is, as a dry powder obtained by freeze-drying. In that form it is stable and does not require a cold chain during transport: shipping does not compromise the material. On receipt, it is kept refrigerated between 2 and 8 °C, protected from light.

For the GHK-Cu strengths, a reference reconstitution would be: el vial de 50 mg with 2 mL of bacteriostatic water queda a 25 mg/mL; el vial de 100 mg with 3 mL of bacteriostatic water queda a 33.33 mg/mL. The volume is not fixed — more water gives a more dilute solution with a larger measuring margin; less water concentrates it — and the choice depends on the working protocol.

The diluent is added slowly along the wall of the vial, never directly onto the powder, and left to rest for five to ten minutes. The vial is swirled gently until fully dissolved; shaking it hard subjects the peptide to mechanical stress that can denature it. Once reconstituted, it is kept refrigerated and is not frozen.

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative, which allows multiple draws from the same vial. Plain sterile water has no preservative and is only suitable for immediate use. Signs of degradation to watch for: cloudy solution, visible particles, change in color or odor.

Frequently asked questions

Why does GHK-Cu have a different FDA status depending on the route?

Because the agency evaluated the routes separately. In the September 29, 2023 update, topical use was placed in Category 1 with limitations, while the injectable routes were flagged in Category 2. Topical evidence is not automatically extrapolated to the injectable route.

Is the copper in the complex relevant?

Yes, it is an essential part of the compound. The tripeptide acts as a carrier of the copper ion to enzymes that depend on it, such as lysyl oxidase, which is involved in the cross-linking of collagen and elastin. Without copper, the GHK tripeptide is not the same compound.

Is GHK-Cu fit for human consumption?

No. It is sold exclusively as a research-use-only (RUO) reagent, labeled as not fit for human consumption. It has no COFEPRIS sanitary registration and no approval from any other agency for human or veterinary use.
