What is BPC-157?

BPC-157 (Body Protection Compound 157) is a synthetic pentadecapeptide, that is, a chain of fifteen amino acids, with the sequence GEPPPGKPADDAGLV. It corresponds to a partial fragment of a protective protein found in human gastric juice, and it is probably the tissue-repair peptide with the most accumulated preclinical literature.

It was characterized in the early 1990s by the group of Predrag Sikirić and Sven Seiwerth at the University of Zagreb, Croatia, as part of a line of work on gastric cytoprotection. Its most distinctive chemical trait is stability: it has no disulfide bridges or post-translational modifications, and it withstands the acidic environment of the stomach without degrading, something uncommon among peptides that made it possible to study it both systemically and orally in animal models.

In terms of chemical identity, it is classified as Synthetic pentadecapeptide, its CAS number is 137525-51-0, its molecular weight is 1419.53 g/mol. These identifiers are what allow the material received to be checked against the batch Certificate of Analysis.

Mechanism studied in preclinical research

The best-documented pathway is the angiogenic one. In 2017, Hsieh and colleagues reported in Journal of Molecular Medicine that BPC-157 increases the expression of vascular endothelial growth factor receptor 2 (VEGFR2) and promotes its cellular internalization, activating the VEGFR2-Akt-eNOS pathway in cascade. The relevant methodological detail is that the effect was observed on the receptor VEGFR2 and not on the ligand VEGF-A, and that it was blocked by inhibiting endocytosis with dynasore, which suggests that receptor internalization is part of the mechanism rather than a side effect.

A later study by the same group, published in Scientific Reports in 2020, described a second related route: BPC-157 promotes the phosphorylation of Src and caveolin-1, which releases endothelial nitric oxide synthase (eNOS) from its inhibitory binding to caveolin-1 and increases nitric oxide production. In isolated rat aorta it produced concentration-dependent vasodilation that disappeared when the endothelium was removed and was inhibited by L-NAME or hemoglobin, confirming that the effect is mediated by nitric oxide.

In tendon injury models, involvement of the FAK-paxillin pathway in fibroblasts has also been described, a pathway involved in cell migration and adhesion to the extracellular matrix. Taken together, the literature does not describe a single receptor: it describes a compound that modulates several converging pathways in vascular and tissue repair, which has been proposed as the explanation for its appearance in models of very different tissues.

What does the evidence on BPC-157 show, and where does it end?

Precision matters here, because this is where most pages about BPC-157 exaggerate. The preclinical base is genuinely broad: more than two hundred published studies over three decades, on intestine, tendon, ligament, muscle, bone, cornea, blood vessels and the nervous system, with an unusual consistency of results.

That same consistency is the point the critical literature highlights. A 2025 review in Pharmaceuticals (Józwiak and colleagues) documented that more than 80% of the records indexed under “BPC 157” are linked to the original Zagreb group, and that the experiments routinely use single doses, with no data on higher, repeated or prolonged exposures. Sikirić's group published a reply defending the compound's profile, and there is a counter-reply: it is an open, documented scientific debate, not a forum controversy.

As for human data, the short answer is that there are none. A 2025 systematic review in HSS Journal (Vasireddi, Hahamyan, Salata and colleagues) synthesized 36 studies published between 1993 and June 2024 —35 preclinical animal studies and one uncontrolled chart review— and documented zero completed controlled clinical trials, a plasma half-life of under 30 minutes and an absence of clinical safety data in humans.

There is also a risk consideration that the literature raises explicitly and that should not be omitted: a compound that increases the expression and activation of VEGFR2 acts on the same receptor that drives tumor vascularization. The 2025 reviews note that there are no in vivo data showing that BPC-157 inhibits tumor progression, and they warn about its pro-angiogenic profile in the presence of subclinical or undiagnosed neoplasms. It is an open question, not resolved experimentally.

How does it differ from related compounds?

BPC-157 is frequently studied alongside other repair compounds, and the difference between them is not one of potency but of mechanism and scope of action.

Handling, reconstitution and storage

The vial is delivered lyophilized, that is, as a dry powder obtained by freeze-drying. In that form it is stable and does not require a cold chain during transport: shipping does not compromise the material. On receipt, it is kept refrigerated between 2 and 8 °C, protected from light.

For the BPC-157 strengths, a reference reconstitution would be: el vial de 10 mg with 1 mL of bacteriostatic water queda a 10 mg/mL; el vial de 20 mg with 2 mL of bacteriostatic water queda a 10 mg/mL. The volume is not fixed — more water gives a more dilute solution with a larger measuring margin; less water concentrates it — and the choice depends on the working protocol.

The diluent is added slowly along the wall of the vial, never directly onto the powder, and left to rest for five to ten minutes. The vial is swirled gently until fully dissolved; shaking it hard subjects the peptide to mechanical stress that can denature it. Once reconstituted, it is kept refrigerated and is not frozen.

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative, which allows multiple draws from the same vial. Plain sterile water has no preservative and is only suitable for immediate use. Signs of degradation to watch for: cloudy solution, visible particles, change in color or odor.

Frequently asked questions

Is BPC-157 approved in any country?

No. No regulatory agency has approved it as a medicine. In the United States it was placed in Category 2 of the 503A list on September 29, 2023, removed from that category in April 2026 and reviewed by the FDA advisory committee in July 2026, but none of those steps amounts to an approval.

Why is it said to be stable in an acidic environment?

Because it contains no disulfide bridges or post-translational modifications, which are the points where other peptides usually degrade. That stability made it possible to study it orally in animal models, something unfeasible with labile peptides such as insulin or native GLP-1.

Are there studies in humans?

There are no completed controlled clinical trials. The 2025 systematic review in HSS Journal identified 35 preclinical animal studies and a single chart review without a control group, between 1993 and June 2024.

Is BPC-157 fit for human consumption?

No. It is sold exclusively as a research-use-only (RUO) reagent, labeled as not fit for human consumption. It has no COFEPRIS sanitary registration and no approval from any other agency for human or veterinary use.
