What is TB-500?

TB-500 is an acetylated heptapeptide with the sequence Ac-LKKTETQ, which corresponds to the actin-binding motif of thymosin beta-4. Precision matters: TB-500 is not thymosin beta-4, but the seven-amino-acid fragment described as responsible for much of its activity.

Thymosin beta-4 is a natural 43-amino-acid protein, present in virtually all body tissues and fluids, whose main function is to sequester G-actin and regulate cytoskeletal polymerization. TB-500 was developed as a shortened synthetic version of that active domain, easier to produce and more stable.

In terms of chemical identity, it is classified as Acetylated heptapeptide, its CAS number is 885340-08-9, its molecular weight is 794.90 g/mol. These identifiers are what allow the material received to be checked against the batch Certificate of Analysis.

Mechanism studied in preclinical research

The described mechanism starts with binding to G-actin. By sequestering actin monomers, thymosin beta-4 and its active fragment modulate the balance between free and polymerized actin, which is what governs cell migration. In repair models, that modulation is associated with greater mobility of endothelial cells and keratinocytes toward the injury site.

The angiogenic and remodeling lines described in the literature derive from that migratory capacity: formation of vascular structures and reorganization of the extracellular matrix. Unlike BPC-157, whose literature describes markedly local effects, TB-500 has been studied with a more systemic distribution profile, which explains why both appear combined in research designs on recovery.

What does the evidence on TB-500 show, and where does it end?

The evidence on full-length thymosin beta-4 is considerably stronger than that on the TB-500 fragment, and they should not be confused when reading the literature: much of what is cited in support of TB-500 actually corresponds to studies with the whole protein.

Thymosin beta-4 reached human clinical trials for ophthalmological and dermatological indications. TB-500 as such has no published controlled clinical trials and is not approved by any regulatory agency.

In the United States it was placed in Category 2 of the 503A list on September 29, 2023 under the name “Thymosin Beta-4, Fragment (LKKTETQ)”, removed from that category in April 2026 and reviewed by the FDA advisory committee on July 23, 2026. As in the other cases, those moves are steps in an administrative procedure on compounding, not approvals.

Handling, reconstitution and storage

The vial is delivered lyophilized, that is, as a dry powder obtained by freeze-drying. In that form it is stable and does not require a cold chain during transport: shipping does not compromise the material. On receipt, it is kept refrigerated between 2 and 8 °C, protected from light.

For the TB-500 strengths, a reference reconstitution would be: el vial de 10 mg with 1 mL of bacteriostatic water queda a 10 mg/mL. The volume is not fixed — more water gives a more dilute solution with a larger measuring margin; less water concentrates it — and the choice depends on the working protocol.

The diluent is added slowly along the wall of the vial, never directly onto the powder, and left to rest for five to ten minutes. The vial is swirled gently until fully dissolved; shaking it hard subjects the peptide to mechanical stress that can denature it. Once reconstituted, it is kept refrigerated and is not frozen.

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative, which allows multiple draws from the same vial. Plain sterile water has no preservative and is only suitable for immediate use. Signs of degradation to watch for: cloudy solution, visible particles, change in color or odor.

Are TB-500 and thymosin beta-4 the same thing?

No. Thymosin beta-4 is a 43-amino-acid protein; TB-500 is the acetylated seven-amino-acid fragment (Ac-LKKTETQ) corresponding to its actin-binding motif. The distinction matters when reading the literature, because many studies cited in support of TB-500 were done with the full protein.

Is TB-500 fit for human consumption?

No. It is sold exclusively as a research-use-only (RUO) reagent, labeled as not fit for human consumption. It has no COFEPRIS sanitary registration and no approval from any other agency for human or veterinary use.
