What is Tirzepatide?

Tirzepatide is a synthetic 39-amino-acid peptide that acts as a dual agonist: on the glucose-dependent insulinotropic polypeptide (GIP) receptor and on the GLP-1 receptor. It is the first molecule of its class to reach regulatory approval.

Mechanism studied in preclinical research

The interest of dual agonism lies in the fact that GIP and GLP-1 are the two main incretin hormones, and their receptors have partially different distributions and effects. Joint activation produces a metabolic profile that neither reproduces on its own.

Structurally it derives from the GIP sequence with modifications that also give it affinity for the GLP-1 receptor, and it incorporates fatty-acid acylation to prolong its half-life through albumin binding, following the same logic as semaglutide.

The diluent is added slowly along the wall of the vial, never directly onto the powder, and left to rest for five to ten minutes. The vial is swirled gently until fully dissolved; shaking it hard subjects the peptide to mechanical stress that can denature it. Once reconstituted, it is kept refrigerated and is not frozen.

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative, which allows multiple draws from the same vial. Plain sterile water has no preservative and is only suitable for immediate use. Signs of degradation to watch for: cloudy solution, visible particles, change in color or odor.
